The Flow Cytometry facility is located in room 2020, Small Animal Hospital (SAH) Building. The facility operator is Dr. Roger Smith III (rosmith@cvm.tamu.edu), 1-979-845-5167. For more information, or to arrange assays, contact Dr. Smith.

Instrumentation

      Becton-Dickinson FACSCalibur, with sorting option.

Software

      CellQuest (Becton-Dickinson)
      ModFit LT (Verity)

Capabilities

Detection of the following fluorochromes:

      Fluorescein and analogs
      R-phycoerythrin
      R-phycoerythrin&shypcyanine 5 (PE-Cy5) tandem dye
      Peridinin chlorophyll protein (PerCP)
      Propidium iodide (PI)
      Doxorubicin (adriamycin)
      Rhodamine 123

Simultaneous detection of up to 3 fluorochromes, e.g.

      Fluorescein,
      R-phycoerythrin, and
Either PE-Cy5 tandem dye or PerCP.

Sorting of cells, in small numbers, for subsequent analysis or culture, e.g.

      Cytology,
      PCR or RT-PCR, or
Isolation of particular cells for subsequent expansion in culture.

  Assays supported

Immunofluorescence (up to 3-color; surface and intracellular antigens)

DNA and cell cycle content analysis (using ModFit LT software)

      Phagocytosis of fluorescent beads

Functional expression of p-glycoprotein (using doxorubicin)
